How can a Mayor resign? is there any option for this ?

There is no in-game tool for resignation from office. The only thing you can do is get a majority of the voters to vote for your opponent.

What would happen if the current mayor untrained politician and there was a running mate?

Well im the running mate in our city and our mayor dropped novice politician and hes still mayor
