if 2 guilds share a city and the total number of people between both are over 55 is it possible to get a shuttleport? Or does one of the guilds need to have 55 alone?

Guild membership doesn't matter, just total number of citizens. If you are 55 citizens and can reach Rank 4, then you can have a shuttleport.

Just be aware that the planet cap of rank 4 cities could've been reached. There's a capon the number of cities on a planet of the different city ranks.
