I heard you could have 2 specializations in a city if you have a Master Politician and are a Metropolis, is this true?

No, the specializations that where available at master politician did consist of a combination of medical center and entertainment district or research and manufacturing center. However this was pulled due to buggyness. It will be reinstated eventually, or alted out.
