Does items in backpacks count now? when you put them on a vendor? So let's say you have 50 items in your backpack and put them on the vendor, will it count for 50 items?

I think so...When I checked my storage vendor I had about 40 items (only filled backpacks). But when i checked my vendors status it said that I had over 900 items for sale (99999999 of course).

Yes, whatever you have in the pack counts against your item total. The bag does not count. I put 12 items in a bag and put it on a vendor that had 108 items on it. When I put it on the vendor, the item total went to 120. So the bag doesnt count, but the items do.
