Im currently TKM/Master Brawler...dropping doctor..and pickin up some fencer Defense.
My question is..should I go
4-0-0-0
or
0-0-4-0
??
and will I have to have a Fencer weapon equiped for these defenses to work?

Depends on the defence you are looking at getting.
If you get the Melee Defence line it will stack up to whatever the cross class cap is. I'm still not sure what it is as I'm Master Fencer TKA 4004 and I don't notice any difference from before the cap was put into place.
The Defence vs state changes add regardless as well I think but I've not seen any metrics to support this like I have the Melee/Ranged defence staking.
The CoB doesn't add... neither does the mitigation you will still be mitigation 3 while useing fists or VK... but while useing Fencer weapon you will be at Mitigation 0-1-2 respectivly as you went up the line that gives it.
