Do they even work? Can they be used with Artisan drinks if they do work. Yes, I looked.

Casks do work, and will make the stack size 3x what you get with a small glass. They can be used in any drink schematic (even artisan ones like Ruby Bliel or Spice tea).
Barrels "work", in that you can use them in the drink container slot. However, you get the same stack size as if you used a small glass, so they're effectively broken. Why use 300 units of metal and two Master Artisan subcomponents when 10 gemstone will do just as well.

So I take it no one will put their exp. points into quantity if they are using casks? Does it go duration, filling, then quantity for experimenting?

As with most questions on experimentation, the answer is "it depends".
Quantity is always an integer value, which means that there are certain "breakpoints" where the quantity changes. Hitting this breakpoint with a small glass increases the quantity by 1, hitting that point with a cask increases it by 3.
If you're just under to a breakpoint, spending a point (or even two) in Quantity can be useful. More than that is probably not worth it, though.
Most foods' breakpoints are at 20%, 25% or 33%. With good PE/DR resources, you can usually exceed the 20/25 values without spending any experimentation points. Of course, with the drink quantity display bug, you can't see where you're at, so it's tough to figure out whether you hit a breakpoint or not. It's beneficial to just make up some practice stacks (don't use tissues, and stick with small glasses) to see what stack size you get with various levels of quantity experimentation.
