I know how to do it to get 1 match, sometimes I get 2 matched - how is this done please! I don't understand the guide on the forums - can someone explain it laymans terms for me!

Think of the list of the bottom as a string of DNA running vertically from top to bottom. Think of laying the sample you are given - lining it up with the base that you choose. If your sequence is AGGT and you match it to TTTA then 2 bases will pair (the first and last). The first base on the top has to match something on the bottom. The bottom strand doesn't change - it just gets paired up.
I hope this helps you visualize it b/c that's what made me finally figure it out myself.
