If you get several samples from the same mob, and you compare them, you'll see the numbers are higher on the higher quality samples. ie hardiness on a good quality is 300 on a vhq it maybe 500 (these numbers are meaningless just an example).
Where the sample comes from has no effect on which skin you can use it in. So yes, DNA from a wort can be used in a dewback or a rancor.
No, you can not sample directly on a sample. If you start crafting, there is a tab on top for genetics. There are 5 slots in which you insert samples of DNA....they can all be from the same or from different mobs. When you combine them, you get the option to experiment on them.
Given the way the crafting is currently borked, check your samples for fortitude...if the sample has fort 1000, you can only use it in slots 1 and 3 otherwise your final sample will have fort >500 which gives it armor and a CL >100.
Following a tip, I found the hambaba's and kahmurra's on Talus have very low fortitude. You might try samples from these mobs.
