Are items that are sent to the stock room deleted instantly?

No.
They will sit there for about 30 days.
However, there is a vendor inactivity timer that will automatically delete a vendor that has no items on sale if it stays that way for a couple of weeks..

Once I place a vendor with business III, do I need to keep the skill to continue use it?

Yes.

Originally you could place vendors, than drop the merchant skills, but at some point this was changed, is the old way was very be-littleing of the Merchant profession.
